If you talk to her again after reloading the room, often she'll just not give you the 50 rupees. You'll have to reload a second time if you want the money.

sounds similar to the frame perfect talk to her when you place a chicken in the pen, and prevents her from giving the item without a reload. why would that happen if it's 0 chickens?

In my experience she always gives you the prize right away the first time you talk. But the next time you're in the area, if you want the 50r, you might have to talk/reload/talk.
