Like unlocking your computer using your phone's NFC instead of a smartcard ?

This project emulates a smart card on your phone. So if your computer can be unlocked with a smart card, then yes, with the mechanism shown in this project, it is generally possible.

What do you mean by unlocking? Do you mean decrypting the HDD? Or do you mean passing the login screen?

Speaking more specifically, it will probably not work with this project out of the box. In order to realize your plan, you should first find out how the unlocking mechanism works with a real smart card. This should show you, what exactly you need to simulate on the smart phone. In this project, a smart card with an SMIME-certificate is emulated. This will most probably not be what is needed for your unlocking mechanism.
